Here, we report the complete genome sequence (2.92 Mb) of Methanosphaerula palustris E1-9C[superscript T], a methanogen isolated from a minerotrophic fen. This is the first genome report of the Methanosphaerula genus, within the Methanoregulaceae family, in the Methanomicrobiales order. E1-9C[superscript T] relatives are found in a wide range of ecological and geographical settings.

The biological sciences encompass topics considered controversial by the American public, such as evolution and climate change. We believe that the development of climate change education in the biology classroom is better informed by an understanding of the history of the teaching of evolution. A 3.19-Mbp draft genome of the Gram-positive thermophilic iron-reducing Firmicutes isolate from the Peptococcaceae family, Thermincola ferriacetica Z-0001, was assembled at ~100× coverage from 100-bp paired-end Illumina reads. The draft genome contains 3,274 predicted genes (3,187 protein coding genes) and putative multiheme c-type cytochromes.

Cocoamidopropyl betaine (CAPB), which is a biodegradable ampholytic surfactant, has recently been found to dramatically enhance the aerobic digestion of waste activated sludge (WAS) in short-time aerobic digestion (STAD) systems. Therefore, it is important to understand the mechanisms in which CAPB enhances WAS aerobic digestion performance. Results showed that CAPB could dramatically enhance the solubilization of soluble proteins (PN), polysaccharides (PS), nucleic acids (NA) and humic-like substances (HS) in the STAD system within the initial 2 h. The relationship between sequence and binding properties of an aptamer for immunoglobulin E (IgE) was investigated using custom DNA microarrays. Single, double and some triple mutations of the aptamer sequence were created to evaluate the importance of specific base composition on aptamer binding. The majority of the positions in the aptamer sequence were found to be immutable, with changes at these positions resulting in more than a 100-fold decrease in binding affinity. Anode-respiring bacteria (ARB) generate electric current in microbial electrochemical cells (MXCs) by channeling electrons from the oxidation of organic substrates to an electrode. Production of high current densities by monocultures in MXCs has resulted almost exclusively from the activity of Geobacter sulfurreducens, a neutrophilic freshwater Fe(III)-reducing bacterium and the highest-current-producing member documented for the Geobacteraceae family of the Deltaproteobacteria. Here we report high current densities generated by haloalkaliphilic Geoalkalibacter spp., thus broadening the capability for high anode respiration rates by including other genera within the Geobacteraceae. We sequenced and annotated genomes of two haloalkaliphilic Deltaproteobacteria, Geoalkalibacter ferrihydriticus Z-0531[superscript T] (DSM 17813) and Geoalkalibacter subterraneus Red1[superscript T] (DSM 23483). During assembly, we discovered that the DSMZ stock culture of G. subterraneus was contaminated. We reisolated G. subterraneus in axenic culture and redeposited it in DSMZ and JCM.

Syntrophic interactions between organohalide-respiring and fermentative microorganisms are critical for effective bioremediation of halogenated compounds. This work investigated the effect of ammonium concentration (up to 4 g liter[superscript −1] NH4+-N) on trichloroethene-reducing Dehalococcoides mccartyi and Geobacteraceae in microbial communities fed lactate and methanol. We found that production of ethene by D. mccartyi occurred in mineral medium containing ≤2 g liter[superscript −1] NH4+-N and in landfill leachate. For the partial reduction of trichloroethene (TCE) to cis-dichloroethene (cis-DCE) at ≥1 g liter[superscript −1] NH4+-N, organohalide-respiring dynamics shifted from D. mccartyi and Geobacteraceae to mainly D. mccartyi. Butyrate is a common fatty acid produced in important fermentative systems, such as the human/animal gut and other H[subscript 2] production systems. Despite its importance, there is little information on the partnerships between butyrate producers and other bacteria. The objective of this work was to uncover butyrate-producing microbial communities and possible metabolic routes in a controlled fermentation system aimed at butyrate production. The inverse problem associated with electrochemical impedance spectroscopy requiring the solution of a Fredholm integral equation of the first kind is considered. If the underlying physical model is not clearly determined, the inverse problem needs to be solved using a regularized linear least squares problem that is obtained from the discretization of the integral equation. For this system, it is shown that the model error can be made negligible by a change of variables and by extending the effective range of quadrature.
