Telomerase is a special reverse transcriptase that extends the linear chromosome termini in eukaryotes. Telomerase is also a unique ribonucleoprotein complex which is composed of the protein component called Telomerase Reverse Transcriptase (TERT) and a telomerase RNA component (TR). The enzyme from most vertebrate species is able to utilize a short template sequence within TR to synthesize a long stretch of telomeric DNA, an ability termed "repeat addition processivity". By using human telomerase reconstituted both in vitro (Rabbit Reticulocyte Lysate) and in vivo (293FT cells), I have demonstrated that a conserved motif in the reverse transcriptase domain of the telomerase …

Contemporary theories of trauma identify the creation of a coherent trauma narrative and therapeutic exposure to trauma memories as potential recovery mechanisms. These factors are often inherent to the disclosure process, resulting in a parallel theoretical framework for experimental research that conceptualizes disclosure as a therapeutic intervention. The present investigation examined the moderational impact of disclosure following trauma on the link between trauma severity and symptoms of Posttraumatic Stress Disorder (PTSD). This thesis examines the modeling, analysis, and control system design issues for scramjet powered hypersonic vehicles. A nonlinear three degrees of freedom longitudinal model which includes aero-propulsion-elasticity effects was used for all analysis. This model is based upon classical compressible flow and Euler-Bernouli structural concepts. Higher fidelity computational fluid dynamics and finite elementmethods are needed formore precise intermediate and final evaluations. Since scholars first turned their attention to the subject some eighty years ago, one major area of contention in the study of Anglo-Saxon wills has been the function of the written will within Anglo-Saxon culture. Verbal agreements, formalized through oral ceremonies and symbolic actions, were recognized as legally binding; however, many of these agreements were also recorded in writing. Many scholars argue that the written document was superfluous;oral ceremonies were written down only in the case of memory failure and the documents themselves had no real performative function. "Globesity," as defined by the World Health Organization, describes obesity as a pandemic affecting at least 400 million people worldwide. The prevalence of obesity is higher among women than men; and in non-Hispanic black and Hispanic populations. Obesity has been significantly associated with increased all-cause mortality, and mortality from cardiovascular disease, obesity-related cancers, diabetes and kidney disease. Current strategies to curb obesity rates often use an ecological approach, suggesting three main factors: biological, behavioral, and environmental. Prognostics and health management (PHM) is a method that permits the reliability of a system to be evaluated in its actual application conditions. This work involved developing a robust system to determine the advent of failure. Using the data from the PHM experiment, a model was developed to estimate the prognostic features and build a condition based system based on measured prognostics. To enable prognostics, a framework was developed to extract load parameters required for damage assessment from irregular time-load data. To establish reliable wireless communication links it is critical to devise schemes to mitigate the effects of the fading channel. In this regard, this dissertation analyzes two types of systems: point-to-point, and multiuser systems. The music of Johann Sebastian Bach has long been used for keyboard percussion pedagogy and performance. The cello suites (BWV 1007-1012), in particular, are popular choices for marimbists. As with many transcriptions for marimba, performers are challenged to transfer Bach's musical genius onto an instrument whose timbre, range, mode of execution and acoustic properties are distinctly different from the original. To date, there is no concise and relevant edition of the suites for study and performance at the marimba. Many methods have been proposed to estimate power system small signal stability, for either analysis or control, through identification of modal frequencies and their damping levels. Generally, estimation methods have been employed to assess small signal stability from collected field measurements. The existing compact models can reproduce the characteristics of MOSFETs in the temperature range of -40oC to 125oC. Some applications require circuits to operate over a wide temperature range consisting of temperatures below the specified range of existing compact models, requiring wide temperature range compact models for the design of such circuits. ABSTRACT This study explored the functions of dialogue in emerging adults' moral turning points. Through purposive sampling, the researcher interviewed 10 emerging adults between 25 and 30 years old about experiences of turning point conversations during the years of 18 and 25. This study employed constant comparative and grounded theory methodologies to analyze messages reported in memorable conversations during this period. Results indicated that dialogue functioned to educate, disturb, and maintain emerging adults' moral perception during this period of moral reorientation. The purpose of this study was to explore current pedagogical approaches of undergraduate directing curricula in selected U.S. institutions of higher learning. Building on the work of Clifford Hamar and Anne Fliotsos, the thesis builds a foundation for further study of contemporary directing pedagogy. Fourteen course syllabi were collected voluntarily from members of The Association for Theatre in Higher Education (ATHE) and served at the primary source material. They were interpreted and analyzed qualitatively for components that identified the methods and philosophies of the instructor and/or institution. The global transport and deposition of anthropogenic nitrogen (N) to downwind ecosystems are significant and continue to increase. Indeed, atmospheric deposition can be a significant source of N to many watersheds, including those in remote, unpopulated areas. Bacterial denitrification in lake sediments may ameliorate the effects of N loading by converting nitrate (NO3-) to N2 gas. Denitrification also produces nitrous oxide (N2O), a potent greenhouse gas. Thousands of high-resolution images are generated each day. Segmenting, classifying, and analyzing the contents of these images are the key steps in image understanding. This thesis focuses on image segmentation and classification and its applications in synthetic, texture, natural, biomedical, and industrial images. The bony pelvis is a pivotal component of the locomotor system, as it links the hindlimb with the trunk and serves as anchorage for the primary propulsive musculature. Its shape is therefore expected to be adapted to the biomechanical demands of habitual locomotor behavior. Codeswitching, or the bilingual practice of switching between two languages, is a frequently misunderstood phenomenon in many fields, including education. Given the growing number of bilingual students and English Language Learners in U.S. schools, it is imperative that the field of education be informed by current research in bilingualism and language acquisition, including codeswitching. In the past, it has been assumed that measurement and predictive invariance are consistent so that if one form of invariance holds the other form should also hold. In the United States, recreational female golfers give up golf at twice the rate of recreational male golfers. This study explored the causal attributions of 240 recreational golfers after a practice session at a public golf facility. Attributions can be adaptive or maladaptive after a performance and can influence subsequent motivation to engage in a similar task again. It was hypothesized that male and female golfers would make significantly different attributions for their performance and that female golfers' attributions would be maladaptive. The Resource Description Framework (RDF) is a specification that aims to support the conceptual modeling of metadata or information about resources in the form of a directed graph composed of triples of knowledge (facts). RDF also provides mechanisms to encode meta-information (such as source, trust, and certainty) about facts already existing in a knowledge base through a process called reification. An investigation of phase noise in amplifier and voltage-controller oscillator (VCO) circuits was conducted to show that active direct-current (DC) bias techniques exhibit lower phase noise performance than traditional resistive DC bias techniques. Low-frequency high-gain amplifiers like those found in audio applications exhibit much better 1/f phase noise performance and can be used to bias amplifier or VCO circuits that work at much higher frequencies to reduce the phase modulation caused by higher frequency devices. The nature and correlates of emerging internalizing symptoms in young children are largely unknown. Maternal factors such as psychological symptoms and detached parenting style have been found to be present in children with anxiety and depression. Further, child attentional control in task completion has been associated with difficulty related to internalizing problems. Stress responses play a central role in the development of psychopathology. Coping efforts, one subset of stress responses, have been shown to influence the relations between stress and adjustment. Graph coloring is about allocating resources that can be shared except where there are certain pairwise conflicts between recipients. The simplest coloring algorithm that attempts to conserve resources is called first fit. Interval graphs are used in models for scheduling (in computer science and operations research) and in biochemistry for one-dimensional molecules such as genetic material. Biology textbooks are everybody's business. In accepting the view that texts are created with specific social goals in mind, I examined 127 twentieth-century high school biology textbooks for representations of animal development. Paragraphs and visual representations were coded and placed in one of four scientific literacy categories: descriptive, investigative, nature of science, and human embryos, technology, and society (HETS). I then interpreted how embryos and fetuses have been socially constructed for students. I also examined the use of Haeckel's embryo drawings to support recapitulation and evolutionary theory. The Wechsler Intelligence Scale for Children-Fourth Edition (WISC-IV) General Abilities Index (GAI) and Cognitive Proficiency Index (CPI) have been advanced as possible diagnostic markers of Attention-Deficit Hyperactivity Disorder (ADHD). Diagnostic utility statistics were used to test the ability of GAI-CPI difference scores to identify children with ADHD. For this thesis a Monte Carlo simulation was conducted to investigate the robustness of three latent interaction modeling approaches (constrained product indicator, generalized appended product indicator (GAPI), and latent moderated structural equations (LMS)) under high degrees of nonnormality of the exogenous indicators, which have not been investigated in previous literature. Results showed that the constrained product indicator and LMS approaches yielded biased estimates of the interaction effect when the exogenous indicators were highly nonnormal. Fluctuating flow releases on regulated rivers destabilize downstream riverbanks, causing unintended, unnatural, and uncontrolled geomorphologic changes. These flow releases, usually a result of upstream hydroelectric dam operations, create manmade tidal effects that cause significant environmental damage; harm fish, vegetation, mammal, and avian habitats; and destroy riverbank camping and boating areas. This work focuses on rivers regulated by hydroelectric dams and have banks formed by sediment processes. For these systems, bank failures can be reduced, but not eliminated, by modifying flow release schedules. Early hominins present an unusual pattern of sexual dimorphism. On one hand, the canine teeth of these species are weakly size-dimorphic, vertically short, and nonhoning, suggesting a social system characterized by infrequent, low-intensity intermale competition and monogamous pair-bonding. This thesis looks at the 1842 Supreme Court ruling of Prigg v. Pennsylvania, the events leading up to this case, and the subsequent legislative fallout from the decision. The Supreme Court rendered this ruling in an effort to clear up confusion regarding the conflict between state and federal law with regard to fugitive slave recovery. Instead, the ambiguities contained within the ruling further complicated the issue of fugitive slave recovery. This complication commenced when certain state legislatures exploited an inadvertent loophole contained in the ruling. The aim of this project is an exploration of health literacy as found in the papal encyclical Humanae Vitae. The rhetoric of the Catholic Church clearly demonstrates its creation and promotion of moral authority over the health practices of the faithful. As such, the encyclical illustrates the means by which Catholic conscience dictates corporal existence. Through its denunciation of the evolving social mores of the 1960s, its condemnation of contraception, and its encouragement in the reception of natural law, the document offers the merits of Catholic marriage as guiding principles beneficial to all good men. As the world energy demand increases, semiconductor devices with high energy conversion efficiency become more and more desirable. The energy conversion consists of two distinct processes, namely energy generation and usage. In this dissertation, novel multi-junction solar cells and light emitting diodes (LEDs) are proposed and studied for high energy conversion efficiency in both processes, respectively. The first half of this dissertation discusses the practically achievable energy conversion efficiency limit of solar cells. Spatial uncertainty refers to unknown error and vagueness in geographic data. It is relevant to land change and urban growth modelers, soil and biome scientists, geological surveyors and others, who must assess thematic maps for similarity, or categorical agreement. In this paper I build upon prior map comparison research, testing the effectiveness of similarity measures on misregistered data. Though several methods compare uncertain thematic maps, few methods have been tested on misregistration. A study was conducted to assess the effects of generational status on various measures of stigmatization, acculturative stress, and perceived social and interpersonal threat within the Mexican heritage population in the Southwest. The role of the fear of stigma by association, regardless of actual experiences of stigmatization, was investigated, including its relationships with acculturative stress, perceived threat, and social distancing. Exploratory analyses indicated that first generation Mexican Americans differed significantly from second generation Mexican Americans on the perception of Mexican nationals as ingroup members, the fear of stigma by association by Americans, and levels of acculturative stress. The Juglandaceae (walnuts, hickories, pecans) has one of the best-documented fossil records in the Northern Hemisphere. The oldest modern genus, Cyclocarya, today restricted to China, first appears in the late Paleocene (57 ma) of North Dakota, USA. Unlike walnuts and pecans that produce edible fruits dispersed by mammals, Cyclocarya fruits are small nutlets surrounded by a prominent circular wing, and are thought to be wind- or water-dispersed. Since the late nineteenth century, the ware has been manufactured for sale, and a small number of individuals continue to produce the pottery today. Over the past forty years, the amount of pottery in museum and private collections has increased dramatically. Peptides offer great promise as targeted affinity ligands, but the space of possible peptide sequences is vast, making experimental identification of lead candidates expensive, difficult, and uncertain. Computational modeling can narrow the search by estimating the affinity and specificity of a given peptide in relation to a predetermined protein target. The predictive performance of computational models of interactions of intermediate-length peptides with proteins can be improved by taking into account the stochastic nature of the encounter and binding dynamics. In this thesis two methodologies have been proposed for evaluating the fault response of analog/RF circuits. These proposed approaches are used to evaluate the response of the faulty circuit in terms of specifications/measurements. Faulty response can be used to evaluate important test metrics like fail probability, fault coverage and yield coverage of given measurements under process variations. However, it is possible for these devices to be exposed to ionizing radiation during various medical therapeutic and diagnostic activities while implanted. This commands that these devices remain fully operational during, and long after, radiation exposure. The present study was motivated by the fact that there are typically fewer females than males advancing in science, and therefore fewer female science instructor role models. The purpose of this study was to evaluate whether five select scales of the MMPI-A (F, Scale 2, A-dep, A-lse, and A-aln) are predictive of a diagnosis of a major depressive episode according to the current DSM-IV-TR criteria. Participants were 90 girls and 58 boys in a clinical psychiatric setting. The study examined two separate hypotheses across the five scales. The RADiation sensitive Field Effect Transistor (RADFET) has been conventionally used to measure radiation dose levels. These dose sensors are calibrated in such a way that a shift in threshold voltage, due to a build-up of oxide-trapped charge, can be used to estimate the radiation dose.
